As I've learned more about 28bio, I'm convinced that they are at the cutting edge of organoid technology for high throughput drug discovery. In particular, their models of PNS organoids can accurately predict neurotoxicity 7.4x better than animal models. Animal models were the best option for decades, but recently, increasing attention has been paid to their inadequacy for predicting disease outcomes in humans. Organoids are more effective (and frankly, more ethical) than animal studies, but they still have a long way to go. Most iPSC derived CNS organoids include basic cell types in the brain (astrocytes and inhibitory/excitatory neurons), but lack comprehensive recapitulation. Microglia for example, migrate from the yolk sac rather than differentiating from progenitors like other cell types in the brain, and as such many organoid models fail to include them, despite their indisputable relevance for disease outcomes. This field is advancing rapidly, and I am excited to see where it will go next, and even more excited to be able to take part in it with my current endeavors using human dorsal forebrain organoids in the Colak Lab at Weill Cornell Medicine.
